MD033 T4 DNA Polymerase

A DNA polymerase that synthesize DNA in the 5'→3’ direction and strong 3'→5' exonuclease activities.

Product Descriptions

T4 DNA Polymerase catalyzes DNA synthesis in the 5'→3' direction on a single-stranded DNA template with primers. This enzyme exhibits strong 3'→5' exonuclease activity and lacks 5'→3' exonuclease activity. Its 3'→5' exonuclease activity is stronger than that of E. coli DNA Polymerase I. This recombinant T4 DNA Polymerase is expressed in and purified from an E. coli strain that carries the T4 DNA Polymerase gene.

Applications

  • Gap filling for 5’ overhang to generate blunt ends.

  • Removal of 3’ overhang to generate blunt ends.

  • Subcloning following single-strand deletion.

  • Synthesis of the second strand for site-directed mutagenesis applications.

Unit Definition

One unit of the enzyme catalyzes the incorporation of 10 nmol of deoxyribonucleotides(dNTPs)into acid insoluble material in 30 minutes at 37°C.

Highlights

  • Filling in 5' overhang to create blunt ends.

  • Removal of 3' overhang to produce blunt ends.

  • Stronger 3'-5' exonuclease activity than DNA polymerase I (E. coli) and Klenow fragment.
