The recombinant T4 DNA ligase is expressed and purified from a E. coli strain that carries the gene. T4 DNA ligase catalyzes the formation of a phosphodiester bond between juxtaposed 5'-phosphate and 3'-hydroxy termini of double-stranded DNA or RNA. T4 DNA ligase will join blunt or cohesive ends as well as repair single-stranded nicks in double-stranded DNA, RNA, or DNA/RNA hybrids.

Unit Definition

One unit is defined as the amount of enzyme required to give 50% ligation of 6 µg of Lambda-HindIII DNA in 30 minutes at 16°C in a total reaction volume of 20 µl.

Superior performance of  Fast T4 DNA Ligase for fast and highly efficient ligation.  A FAM-labeled dA-tailed dsDNA and the Y-adaptor were used to assess the ligation efficiency measured by capillary electrophoresis (CE). Example CE traces shows 90% of ligation efficiency after 15 mins of incubation at 20oC.
